GPCL has validated several technology options related to investigating the role of miRNA expression in biological systems. These include:
- Small RNA extraction - Small RNA has traditionally been regarded as degradation products and many extraction protocols have been developed to minimize "contamination" of total RNA with smaller fragments. Knowledge about the importance of small RNA in cellular function has changed that attitude and several commercial kits have been developed to optimize retention of these molecules. GPCL undertook a detailed study of several of these kits. A poster describing our methods and results is available. Our specimen processing facility can provide RNA extraction using a method appropriate for your study. Please contact GPCL to discuss your options.
- Realtime PCR analysis - GPCL examined the sensitivity and ease of use of two major producers of assays for realtime PCR of miRNA. A poster describing our methods and results is available. Our realtime PCR facility can advise and assist in developing strategies for global or specific examination of miRNA expression via realtime PCR. Assay primer sets are available from a number of vendors including Applied Biosystems, Qiagen, and Exiqon. - Microarray analysis - There are a number of commercial options for examination of miRNA expression profiles via microarray. GPCL has been successful with the Exiqon LNA arrays, however our equipment and skill of our personnel are compatible with any array type on a standard 1 x 3 slide using a dye labels in the Cy3 and Cy5 flourescence range including those by Invitrogen, Agilent, or Ambion.
