iPLEX® Gold Assay
The iPLEX Gold assay combines the benefits of a simple and robust single-base primer extension biochemistry with the sensitivity and accuracy of MALDI-TOF MS detection. Assays can be multiplexed up to 36 SNPs in one individual reaction. The assay design is automated and straight forward, all oligonucleotides are unlabeled and of standard quality.
The iPLEX® Gold assay is based on multiplex PCR followed by a single base primer extension reaction. After the PCR, remaining nucleotides are deactivated by SAP treatment. The single base primer extension step is performed, and the primer extension products are dispensed to an iPLEX SpectroCHIP® for detection and analysis on the MassArray Workstation.

Sample Preparation
Samples must be submitted in 96 well plates with well H12 empty. Genomic DNA should be at 10ng/ul. If the DNA will be submitted at any other concentration, then this must be noted and discussed with the GPCL (labor charges will apply). Please contact the GPCL for instructions for submission of amplified DNA. Minimum sample volume is 30 ul for up to 8 pools. Submit an additional 2 ul for each additional primer pool to be included. Minimum sample size is 380 and minimum sample increment is 384. Samples and primers must be accompanied by iPLEX sample drop off form and can be delivered to our facilyt at 3343 Forbes Avenue, 3rd floor or to our drop off room at S534 Scaife Hall.
Assay Design and Ordering
Primers for each locus within an assay set must be designed together for maximum multiplex efficiency. Primer design software is available and can be accessed by personnel from a researcher’s lab, or by GPCL staff (additional charges apply). Assay design can begin with a list of rs numbers for loci of interest. Three primers are required per locus, 2 amplification primers and a single extension primer for allele detection. Please contact GPCL to confirm primer design and order layout before placing any primer orders.
